DMT Synthesis from Tryptophan

About six months ago I wrote this paper with no intention of posting it. A lot of research went into it, and it's basically a few papers put together, and rewritten to make it flow. Some of the procedures have been modified to make them easier/functional. DMT from this method is really cheap, and if any of you decide to do this, id love to see some pictures.

Synthesis of Benzene:

1kg Sodium Benzoate and 600g Sodium Hydroxide are mixed and added to a paint can with a hole punctured into the lid, and a glass adapter, wrapped with teflon tape, shoved through. This is then heated with a camping stove whilst connected to a simple to a distillation setup. A decarboxylation is carried out. The product is then washed twice with 20ml Water in a separatory funnel. The top layer is then dried over 10g Calcium Chloride for 2 hours. This crude Benzene is then distilled at 79-80 degrees celsius in order to yield 430ml of pure Benzene.


Saturation of Benzene with Hydrogen Chloride:

In a glass dish containing 1L Benzene place an inverted glass funnel into the Benzene, without it touching the bottom of the glass, using a lab stand. Later in the procedure the funnel will suck up Benzene (due to a drop of pressure) and the Benzene will drop below the height of the funnel's lip, causing the Benzene to fall back into the dish, since there would no longer be a seal around the edge of the funnel. Connect the funnel, with vinyl tubing, to the side of an outer luer adapter, that goes into a 1L round bottom flask. This will stop any backflow from going into the reaction flask, and any backflow that overpowers the funnel will be deposited into the round bottom flask. Then connect the top of the outer luer adapter to a 2L erlenmeyer flask that has a vacuum adapter with vinyl tubing. 60g Sodium Chloride is then added to the erlenmeyer flask. Then 30ml concentrated Sulfuric Acid is added to an addition funnel, and inserted into the top of the flask. Use moderate stirring in the receiving dish, and heavy stirring in the reaction flask, then open the valve on the addition funnel to proceed with the reaction. What is left in the dish, and round bottom flask is saturated Benzene.


Synthesis of Tryptamine HCL:

250mg Tryptophan is added to 10g Diphenylmethane in a 25ml round bottom flask, and refluxed under nitrogen until the mixture stops bubbling. Cool the solution in an ice bath until it reaches room temperature, then add 20ml Benzene saturated with Hydrogen Chloride. Filter off the precipitate with a buchner funnel, then wash it with Hexane. Dry the filtrate, and recrystallize it with Ethanol to yield 150mg of pure Tryptamine HCL.



Synthesis Of N, N DMT:

10g Tryptamine HCL, 6.28g Sodium Cyanoborohydride, 400ml Methanol, and 11.76g Acetic Acid are added to a 1L 3 necked flask, and cooled to 0°C under Nitrogen. 4.2g Formaldehyde is added to 125ml Methanol and slowly added to the flask, with an addition funnel, over the period of about an hour with mild stirring. The flask is then stoppered, and allowed to reach room temperature in a ice bath with little ice. Let reaction mixture sit for 60 hours, then basify the solution to a PH of 13-14 with Sodium Hydroxide. Proceed with 5 100ml extractions of DCM, then combine the extractions. Rotovap off the solvent, then dissolve it with Pet. Ether. Heat to a boil, and filter off any undissolved material(make sure to wash the flask with Pet. Ether). Cool the Pet. Ether in the freezer, and filter off the precipitated material, isolating the solution in a beaker. The precipitated material is Tryptamine, which can be used in future reactions. Place the beaker in an ice bath with mild stirring, and slowly add 100 ml (or equal volume of reaction mixture) of Acetic Anhydride over a period of 1 hour. Filter off the precipitate, and rotovap off the Pet. Ether to yield roughly 7.88 grams N, N DMT.


Costs of Lab Equipment and Chemical Supplies:

The total cost of all the lab equipment bought at professional retailers would cost around $5,860, and would perform exceptionally. An alternative to buying from companies such as Fisher Scientific, would be buying from Ebay. Ebay sells usable lab equipment at an astoundingly low price. All of the equipment from ebay would cost around $2,020. To make a batch of DMT it costs a total of $29.6. Each gram costs $3.76 to manufacture at this scale. These prices are based on chemicals sold from Fisher Scientific. It is well advised that this source is used since it is not only relatively inexpensive, but it also has purity standards, unlike sources such as ebay. If you have a greater budget, Sigma Aldrich would be the best bet, as they have the highest purity standards. If you decide to buy from ebay, please be aware that you will need to purify the substances prior to beginning any of the process. Not only will this exponentially increase your yield, but it will also ensure a useable product. If every part of the reaction is pure, then you can ensure that the final product will be pure as well. Impurities in the final product can be dangerous to the health of the consumer, since precursors and unidentifiable contaminants have the potential to be toxic. When it comes to pharmaceuticals, extreme purity is necessary.
